چکیده

While referee #1 express some interest in the study, I am afraid that the other two referees are not persuaded that the analysis is well suited for publication here. Referee #2 finds that further definitive insight into the identity and regulation of the protease responsible for TGFalpha cleavage is needed. Referee #3 while appreciating that the findings extend previous work, s/he is also not persuaded that advance and insight provided is sufficient to consider publication in the EMBO Journal. Given these opinions and as the EMBO Journal only accepts manuscripts which receive an enthusiastic response upon initial review, I am afraid that our conclusion in this case must be a negative one, and I have therefore decided to return the manuscript to you at this point, with the message that we cannot offer to publish it.

LPA-producing enzyme PA-PLA₁α regulates hair follicle development by modulating EGFR signalling.

Recent genetic studies of human hair disorders have suggested a critical role of lysophosphatidic acid (LPA) signalling in hair follicle development, mediated by an LPA-producing enzyme, phosphatidic acid-selective phospholipase A(1)α (PA-PLA(1)α, also known as LIPH), and a recently identified LPA receptor, P2Y5 (also known as LPA(6)). However, the underlying molecular mechanism is unknown. Her...

Enpp2/Autotaxin in Dermal Papilla Precursors is Dispensable for Hair Follicle Morphogenesis

Systematic ablation of previously identified dermal papilla (DP) signature genes in embryonic DP precursors will reveal their functional roles during hair follicle morphogenesis. In this study, we validate Enpp2/Autotaxin as one of the highest expressed signature genes in postnatal DP, and demonstrate specific expression of this lysophosphatidic acid (LPA)-generating enzyme in embryonic dermal ...

MicroRNA-214 controls skin and hair follicle development by modulating the activity of the Wnt pathway

Skin development is governed by complex programs of gene activation and silencing, including microRNA-dependent modulation of gene expression. Here, we show that miR-214 regulates skin morphogenesis and hair follicle (HF) cycling by targeting β-catenin, a key component of the Wnt signaling pathway. miR-214 exhibits differential expression patterns in the skin epithelium, and its inducible overe...

Re-activation of Wnt/β-catenin Signaling Pathway in Hair Follicle Stem Cells in Treatment of Androgenetic Alopecia

Hair loss is a common hair disorder in human population. It affects quality of life and there are ongoing attempts to find permanent treatment for this condition. But, today there is no completely safe and protective treatment for all. Hair follicle stem cells are alive, but quiescence in androgenetic alopecia and are potentially active and can proliferate and differentiate, then regenerate hai...

Gsdma3 regulates hair follicle differentiation via Wnt5a-mediated non-canonical Wnt signaling pathway

Hair follicle is a mini-organ that consists of complex but well-organized structures, which are differentiated from hair follicle progenitor or stem cells. How non-canonical Wnt signaling pathway is involved in regulating hair follicle differentiation remains elusive. Here we showed that Wnt5a regulates hair follicle differentiation through an epithelial-mesenchymal interaction mechanism in mic...
